Materials:
glass slides, bacterial colony or culture, water bottle, inoculating loop, Bunsen Burner,
marker, drying slide rack, metal tongs, microscope, test tube rack
Procedure:
1. Use only a clean glass slide. Draw circles on the underside of the slide with the marker. Label the
slide on the furthest end with the bacterial type and the date.
2. Turn on Bunsen Burner
3. If the bacterial culture is from a liquid media: NO NEED TO ADD WATER TO SLIDE!
– Tap the test tube to suspend the bacterial culture in the tube.
– Set slide on the drying rack
– Take the inoculating loop in your dominant hand and hold as a pencil.
– Pass the loop though the flame several times
– Allow the loop to cool without putting it down onto a surface.
– Hold culture tube in opposite hand removing the top with your thumb and last 2 fingers.
– Pass the neck of the tube through the flame several times
– Dip the sterile loop into the culture
– Pass the neck of the tube through the flame several times.
– Place the top back onto the tube and place tube into the rack
– Place a loopful of the culture onto the circled area on the slide.
– Allow the slide to air dry.
– Pick up the slide with a metal tong on one end.
– Fix the cell to the glass slide by passing it over a flame.
If using a bacterial slant or solid media: Place one loopful of water on the slide in the circled area
before the above procedure of collecting the bacterial sample from the slant. Remember that in
a slant, less is better so only take a small sample of the slant per circled area on the slide and
mix in the water droplet.
